Behind the film: Retatrutide references & context
Evidence cutoff: October 12, 2026. This guide accompanies a two-minute educational film. It distinguishes discovery research, experimentally informed molecular models, clinical evidence, company statements and registered trial locations. WPF does not sell peptides or provide medical advice. No independent human scientific review or institutional endorsement is claimed.
What is retatrutide?
Retatrutide, also identified as LY3437943, is an investigational peptide developed by Lilly. It activates the receptors for GIP, GLP-1 and glucagon. One engineered molecule interacts separately with these different receptors. It is not a mixture of three medicines, a peptide bridging three receptor proteins at once, or a new hormone named “GLP-3.” Three targets do not establish three times the clinical benefit or equal potency at every receptor.
The 2022 discovery paper describes balanced glucagon/GLP-1 receptor activity and greater GIP receptor activity in its experimental pharmacology. Those comparisons depend on the assay and should not be converted into a fixed percentage contribution to human weight loss.
Discovery and the people behind the work
The film credits Tamer Coskun, Shweta Urva, Kyle Sloop and colleagues, reflecting a team effort. The discovery publication lists Lilly affiliations in Indianapolis, United States, and Vienna, Austria. Our original map illustrates those affiliations; it does not identify a single room or claim that every discovery step occurred at one campus.

Subsequent structural researchers and clinical investigators deserve separate credit. An investigator’s affiliation does not establish that they invented the molecule, and the existence of a photograph of a campus does not make it the verified discovery laboratory.
What the molecular sequence actually shows
The moving receptor views were rendered locally from deposited coordinate files:
| Entry | Receptor complex | Film selection |
|---|---|---|
| 8YW3 | Human GLP-1 receptor | Receptor author chain R; peptide author chain P |
| 8YW4 | Human GIP receptor | Receptor author chain R; peptide author chain P |
| 8YW5 | Human glucagon receptor | Receptor author chain R; peptide author chain P |
The coordinate models were fitted to experimental cryo-electron-microscopy data using research constructs. They are not footage of receptors working inside a person. Gold tubes depict the deposited peptide backbone segment; teal tubes depict the receptor backbone. The translucent receptor envelope is an illustrative rendering derived from the atom positions, not a measured membrane boundary or an exact molecular surface calculation. Colors are a visual key, not the intrinsic colors of these molecules.
Other complex components, including G-protein/helper chains, are omitted for clarity. Unresolved portions are not invented or extended into a supposed complete drug structure. Backbone gaps are kept separate. Camera movement applies a rigid rotation and does not deform the molecular coordinates. Smooth interpolation and enlarged backbone tubes help visibility; they do not simulate molecular dynamics, binding speed or receptor activity.

Its institutions include the Shanghai Institute of Materia Medica, Chinese Academy of Sciences, and Fudan University. This team’s structural contribution is distinct from Lilly’s earlier discovery work.
The PDB archive coordinates are available under CC0. See the RCSB PDB usage policy. We created the views ourselves rather than assuming that journal figures or website screenshots have identical reuse rights.
Mechanism: what is known and what remains unresolved
The discovery paper investigated food intake and energy expenditure. The contribution of glucagon receptor activity to increased energy expenditure was demonstrated in obese mice. That is preclinical evidence. It does not establish how much of a particular person’s outcome is caused by the glucagon component or prove an identical effect in every human tissue.
Our metabolic artwork is a conceptual illustration of this research question. It is not an experimentally observed intracellular drug journey, a literal depiction of retatrutide entering a mitochondrion, or a claim about a direct mitochondrial target. Receptor pharmacology, animal mechanisms and human clinical endpoints are separate kinds of evidence.
TRIUMPH-1: the result and the analysis behind it
By the film’s cutoff date, Phase 3 evidence had been published. TRIUMPH-1 randomized 2,339 adults without diabetes and assessed treatment for 80 weeks. The published treatment-regimen/intent-to-treat analysis reported:
| Study arm | Mean body-weight reduction |
|---|---|
| Retatrutide 4 mg | 17.6% |
| Retatrutide 9 mg | 23.7% |
| Retatrutide 12 mg | 25.0% |
| Placebo | 3.9% |
These are group averages from a specified population and analysis. The doses describe study arms and are not instructions for use. They do not guarantee an individual outcome or demonstrate that a product bought outside the study is equivalent.

Why you also see 28.3%
Lilly’s May 21, 2026 announcement reported a different efficacy estimand. For the 12 mg study arm, that analysis yielded 28.3% versus 2.2% with placebo. Its assumptions concern continuing study intervention, with allowed interruptions/modifications, and avoiding prohibited weight-management treatment.
The treatment-regimen analysis addresses the treatment strategy irrespective of adherence and relevant intervening events. These analyses answer different questions. Keep the comparison pairs together:
| Analysis | Retatrutide 12 mg | Matching placebo |
|---|---|---|
| Published treatment-regimen | 25.0% | 3.9% |
| Company-reported efficacy estimand | 28.3% | 2.2% |
Do not combine 28.3% with 3.9%. Do not present a selected extended-follow-up subgroup as the entire randomized population. The film introduces the distinction without treating either result as a personal guarantee.

Evidence history and different populations
The earlier Phase 2 obesity study remains useful historical evidence: NEJM, 2023 · PubMed 37366315 · DOI: 10.1056/NEJMoa2301972.
Other clinical publications involve different populations and follow-up periods. TRIUMPH-2 and TRANSCEND-T2D-1 should not be folded into one undifferentiated “retatrutide result.” A diabetes study, an obesity study without diabetes and a clinical-outcomes study do not establish the same endpoint. No unverified numerical claims from these additional papers are included in the film.
Worldwide research, grounded in the registry
Our map uses the posted locations in the TRIUMPH-1 registry record. Its locations are in Australia, Brazil, Canada, Hungary, India, Mexico, Poland, Puerto Rico, Romania, Spain, the United Kingdom and the United States. These are countries and territories, not twelve sovereign countries. Registry site markers are based on posted geographic coordinates, which may represent a city rather than a precise building.
| Study | Last posted update in downloaded record | Recorded status | Countries/territories listed |
|---|---|---|---|
| TRIUMPH-1 · NCT05929066 | August 21, 2026 | Completed | Australia, Brazil, Canada, Hungary, India, Mexico, Poland, Puerto Rico, Romania, Spain, UK, US |
| TRIUMPH-2 · NCT05929079 | July 30, 2026 | Completed | Argentina, Australia, Brazil, India, Mexico, Romania, Spain, US |
| TRANSCEND-T2D-1 · NCT06354660 | March 5, 2026 | Completed | India, Mexico, Puerto Rico, US |
These examples span the Americas, Europe, Asia and Australia. They do not establish equal representation, enrollment on every continent, current recruitment or worldwide access. We do not infer absence of all research in a region from the absence of that region in these three records. Trial authorization differs from a medicine’s marketing approval.
Safety and availability
Gastrointestinal adverse effects are common in the reported research, and safety assessment continues. Company reports also discuss altered skin sensations and treatment discontinuation. A complete safety discussion requires the study, dose, population and denominator; the short film does not invent universal rates or import another medicine’s approved warnings.
Lilly’s information updated in September 2026 identifies retatrutide as investigational and not approved by any regulatory agency. This is attributed to the developer rather than presented as an independently completed review of every jurisdiction. Lilly: What to know about retatrutide.
Trial evidence does not verify the identity, potency, sterility, provenance or safety of products sold under the same name outside controlled trials. The film offers no purchasing route, self-administration advice, commercial price or promised approval date.

Captions, updates and corrections
English SRT/VTT files and the transcript are provided separately. Caption timing is estimated within each measured narration segment; it is not a word-level forced alignment. Captions are optional, not burned into the film.
Later findings can update this guide while preserving the recorded film’s evidence cutoff. Corrections or useful primary sources can be sent to admin@worldpeptidefoundation.org. Scientific studies and self-reported observations remain clearly distinct. An expert opinion, source verification and independent scientific assessment are not interchangeable.

One peptide. Three receptors. Retatrutide extends the story beyond the incretin medicines already in clinical use.
Lilly researchers, including Tamer Coskun, Shweta Urva, Kyle Sloop and colleagues, published its discovery and early human proof-of-concept work in twenty twenty-two.
Retatrutide activates GLP-one, GIP and glucagon receptors. These are separate interactions of one engineered molecule, not a mixture of three drugs. Deposited structural models help explain how the peptide is recognized by each receptor.
Adding glucagon activity explores energy use as well as food intake. That mechanism was demonstrated in preclinical models; its contribution in humans is not fully resolved.
By October twenty twenty-six, peer-reviewed Phase Three results were available. In TRIUMPH-one, adults without diabetes received retatrutide or placebo for eighty weeks. The published treatment-regimen analysis reported average weight loss of twenty-five percent with the twelve-milligram study dose, versus three point nine percent with placebo. The often-quoted twenty-eight point three percent uses a different analysis. Neither is an individual guarantee.
The research spans multiple continents. Registered trial locations include countries in the Americas, Europe, Asia and Australia; that does not establish worldwide access or equal representation.
Gastrointestinal effects are common, and safety assessment continues. Retatrutide remains investigational. Trial evidence does not verify products sold under the same name.
